The aim of this proposal is to predict the 3D structure of the GLP-1 receptor bound to GLP-1 ligand using the newly developed artificial intelligence algorithm AlphaFold 3.Through a pilot study, we will verify whether AlphaFold 3 can effectively simulate the structure of the GLP-1 receptor bound to ligands and G proteins, which will serve as a basis for the design of next-generation GLP-1 agonists.Successful production of the predicted structures will serve as an important basis for the subsequent rational design of high-performance, low-cost GLP-1 drugs

The aim of this proposal is to systematically investigate the various factors affecting hormone secretion in the gut, including nutrients, drugs and other endocrine signals, using a split-perfusion organ model. Through this physiological model, which can precisely control the stimulation conditions while preserving the structure and function of the intestine, this study will reveal the regulatory mechanisms of intestinal hormones such as GLP-1 and GIP, laying a key foundation for the development of non-surgically induced hormone release therapeutic strategies in the future, and advancing the development of novel therapeutic treatments for type 2 diabetes mellitus and obesity.

To fund a series of pilot projects on LNP development for nucleic acid delivery. Data generated from successful pilot studies will be leveraged in larger grant applications in order to fund one or more comprehensive and large-scale research programs on rational design and therapeutic uses of LNP systems.
